Problem Statement # 2:
Sustained Drug Delivery for Glioblastoma Multiforme
Glioblastoma multiforme (GBM) is a particularly devastating cancer, the most
common and most aggressive malignant primary
ain tumor. It has a high
incidence of recu
ence, so even with the best, multimodal treatment average life
expectancy is only 15 months. The only significant change in the treatment of GBM
over the past two decades was the development of an intracranial, implantable drug
delivery device, the Gliadel® wafer, which is used only during the initial surgical
intervention. While Gliadel® was at first well received, it has had poor success and
has not been popularly adopted due to the rapid initial drug delivery, and inability
to follow-up with additional delivery upon tumor recu
ence. Your task is to develop
a sustained drug delivery vehicle capable of being reloaded in the incidence of
tumor recu
ence. This could be a completely novel device or a fundamental
modification of an existing device or procedure. Your research proposal is limited to
5 pages (not including figures, tables, and references) and should include a
ief
ackground of the disease and risks/limitations associated with cu
ent treatment
options, a detailed description of your proposed solution with a justification for any
assumptions, and an analysis of the biocompatibility and feasibility of the device.
The description of your proposed solution should also include details on the
therapeutic agent being released to treat the tumor, how the therapeutic agent will
effectively treat the tumor, and the release kinetics of the delivery vehicle.
