Discuss some of the physical and chemical factors involved in the composition, and in the 
preparation, of a culture medium: Nutrient ingredients, pH and buffering, Heat (to 
reconstitute), Heat (to sterilize), other. 
Culturing of microbes may be regarded as an initial step required for the purpose of study 
of their morphology and identification. During culturing, it’s quite necessary to provide proper 
nutrition and atmosphere to microbes so that, they could grow and manipulate in a significant 
mode (Davis et al., 1973). 
Therefore; an artificial medium must provide all the nutritional components which are 
received by microorganisms in their natural habitat. Though, requirement of nutrients may vary 
according to the microorganism being cultured but, usually a medium consists of water, carbon, 
nitrogen & energy source, trace elements and few growth factors for instance;
