Question #1: (10 Marks)

The plant-derived terpene natural product ent-kaurenoic acid (shown below) has shown
promising bioactivity.

Experiments conducted by Adriana Lopes and co-authors (Phytochemistry Letters 2016, 18,
162-167 — available in UMLearn) established the biosynthetic pathway of ent-kaurenoic acid
using 13C–labelled glucose.

Please describe in your own words, the experiments that demonstrated the
biosynthesis of ent-kaurenoic acid in Smallanthus sonchifolius.

Feel free to include as many figures and schemes as you need to explain the results —
BUT: they must be original drawings (not copied directly from the source paper).

Feel free to consult any other source material that helps you describe the experiments
— but please be certain to add them as a reference to your assignment submission.

Maximum Length = Two Pages — plus references.

The assignment submission should be completed in MS Word with embedded figures and
structures as part of the two pages. All chemical structures must be drawn in ChemDraw.

Biosynthesis of ent-kaurenoic acid
ent-kaurenoic acid, is a diterpenoid which is biosynthesized by Smallanthus sonchifolius. It is an essential intermediate of many plant secondary metabolites, for example it is involved in the biosynthesis of gi
erellin, a phytohormone. It has been reported earlier that ent-kaurenoic acid possess cytotoxic effects, em
yotoxic effects and genotoxic effects (Lopes et al.; Cavalcanti et al.). It also exhibits anti-bacterial activities. Recently, it was discovered that it possess anti-biofilm activity, hence helps in killing the bacteria. It’s antibacterial activity was better when compared to gentamycin (Lopes et al.). The synthesis of ent-kaurenoic acid was believed to occur through mevalonate (MVA) pathway, which is involved in the synthesis of terpenoids, however, Lopes et al., showed that biosynthesis of ent-kaurenoic acid is done by...
