INDENTIFICATION OF ANALGESIC IN THE MEDICINE USING TLC
ABSTRACT:
Chromatography (chromous = colour) methods are used widely in organic chemistry
laboratories for routine analysis. TLC is generally used to check the purity of a
sample and to analyze the actual components of a mixture and also to follow the
pace of reactions. Analgesics are a category of drugs, which show the general
property of alleviating pain, can in patients. Some examples are acetaminophen,
aspirin, i
ufen, diclofenac, methylsalicylate, caffeine etc. The active ingredient of the
analgesic in the pain formula can be found out using TLC. In this experiment, TLC is
performed on a mixture of analgesics, in a view to find out the exact components
present in it.
INTRODUCTION
Chromatography can be used to separate mixtures of substances into its
components. There are many form of chromatography and they all work with the
same principle. They all have a stationary phase and a mobile phase. The mobile
phase flows through the stationary phase and ca
ies the components of the mixture
with it. Different components travel at different rates. We'll look at the reasons for this
further down the page. Thin layer chromatography is done exactly as it name
denotes - using a thin, uniform layer of silica gel or alumina coated onto a piece of
glass, plastic etc. The silica gel is the stationary phase. The mobile phase is a
suitable liquid solvent or mixture of solvents.
MATERIALS AND METHODS
1. Standardisation of Rf values
For performing TLC, commercially available precoated TLC plates were used. 2%
solutions of acetaminophen, acetanilide, phenacetin, caffeine and salicylamide were
used to perform TLC. The purpose of this activity was to determine and standardise
the Rf values of these compounds. A TLC plate was of approximate size 10cm x 7.6
