These drugs can act as structural analogs, which mean that they are similar in structure to metabolic intermediates which are present naturally. These analogs compete to the take the position of intermediates in various metabolic processes due to their resemblance, but they are different in such a way that they prevent normal metabolism and functioning of a cell. They are bacteriostatic but broad spectrum antibiotics.
The first antimetabolites to be used successfully as chemotherapeutic agents were sulphonamides, discovered by G. Domagk (Miscellaneous Medical Equipment).
