BIOCHEMICAL OXYGEN DEMAND
INTRODUCTION:
Biochemical Oxygen Demand (BOD) is the amount of oxygen needed by bacteria while stabilizing decomposable organic matter under aerobic conditions. The term “decomposable” may be interpreted as meaning that the organic matter can serve as food for the bacteria, and energy is derived from its oxidation (Sawyer and McCarty). The BOD test is a bioassay in which the rate (and extent) of the aerobic degradation of organic matter is assessed in terms of the amount of oxygen consumed during its degradation. In laboratory BOD test was performed to determine the 5-day BOD, ultimate BOD, and the BOD rate constant of the sample by the dilution method (dilution water is not seeded) i.e. to measure the organic strength and the oxygen requirement of the wastewater generated by the West Mobile area.
METHODOLOGY:
At first initial DO level of the sample was measured twice by using the DO probe and average of the results was taken. BOD bottles were labeled. With estimated DF, two BOD bottles were prepared for each day by putting the appropriate volume of undiluted sample into the BOD bottles and then BOD bottles were filled up with dilution water until full. Before incubation 0.16 grams of nitrification inhibitor was added into each BOD bottle. In the next step, each BOD bottle was closed carefully with stopper to exclude air bu
le and contents in the bottle were mixed by inverting the bottle several times. Then the BOD bottles were stored in the 20degree Celsius refrigerator until measurement. Finally, the DO level of two sample were measured and average of results were taken after 24,48, and 72 hours. Because of time constraints the 72 hour reading was taken as BOD5.
For estimation of DF, following steps were taken. At first, ultimate BOD (Lo) was determines by computing the ThOD of the sample (Sucrose) for this lab.
