The Lingo-1 stands for Leucine-rich repeat and Immunoglobulin-like domain containing Nogo receptor-interacting protein-1. The human Lingo-1 protein is the central nervous system (CNS) specific in nature. The crystal structure of the ligand-binding ectodomain of this protein shows the bimodular and kinked structure. It is constituted of leucine-rich repeats (LRR) and immunoglobulin (Ig)-like segments. The structure along with the biophysical investigation of the properties of its solution had revealed the formation of stable tetramers of the molecule in both states. The LRR-Ig composite fold of Lingo-1 drives the association and assembly of Lingo-1 with itself. The major domains of this protein are the N-terminal LRR module (residues 34-64: N-cap; 66-353: LRR motifs; 363-415: LRR C-cap), Ig1 domain (435-493), the stalk region (494-550) and the...
